2、lants and horticulture Abstract In this study, two ornamental plants of cyclamen and ground-covered chrysanthemum were respectively for anther culture. The impact of anther embryoids induction or callus induction by va
3、rious of factors and histological observation were studied. Finally, ISSR of the regenerated plants derived from anther culture of ground-covered chrysanthemum was analized. Specific studies and findings are as follo
4、ws: 1. Flower size and color can be used as judging microspore development stages simply and reliably.In cyclamen, the petals is longer than the calyx 2mm ~ 6mm and its colour is white or pink, the anthers are at late un
5、inucleate stage; in ground-covered chrysanthemum, flower bud diameter is 0.5cm ~ 0.8cm, the center sepal is close, transparent, and the light yellow florets on the outside can be observed, the anthers are at uninucleate
6、stage in the light yellow florets on the outside. 2. In cyclamen anther culture, the best method of sterilization was: the flower buds rinsed with tap water, immersed 30min in 0.2% carbendazim reagent, and transferred to
7、 clean benches washed twice with sterile water. Then soaked 30s in 70% alcohol, rinsed 1 min by sterile water, and sterilized 20 min with 1.5% NaClO. Finally, rinsed 3 times in sterile water, each 1 min. 3.Different geno
8、types was significant difference in anther culture. The highest rate of embryoids was induced in cyclamen ‘Halios 2010’, ‘Halios2160’, and the highest rate of callus was obtained in ground-covered chrysanthemum ‘Beilinh
9、ong’ . 4.Low-temperature pretreated cyclamen ‘Halio2010’ for 48h, the embryoid induction rate is 17.8%, higher than that of others. In ground-covered chrysanthemum, the plant regeneration rate was the highest by low-tem
10、perature pretreated for 72h. 5.Plant growth regulators played an important role in anther culture. The results showed that the rate of embryoids induction was difference at the medium with same composition of plant gro
11、wth regulators between different cultivars in cyclamen. The highest rate of embryoid induction was obtained in medium with 0.02 mg / L NAA +1.0 mg / L 6-BA for ‘Halios 2010’, while in medium with 0.1mg/L NAA +1.0 mg / L
12、 6-BA for ‘Halios 2160’ . In ground-covered chrysanthemum, the highest callus induction was induced in medium with 1.0mg/L 6-BA +1.0 mg/L 2,4-D for ‘Beilinhong’. 6. Sucrose concentration effected cyclamen embryoids ind
13、uction obviously. The results showed that the rate of cyclamen embryoid induced by adding 90 g/L of medium sucrose is 11.2%, 2 times than 60 g/L and 120 g/L sucrose. 7.Activated carbon did not effect of callus inductio
